總結(jié)	HBA2 located on chromosome 16 spans about 30 kb and includes seven loci: 5'- zeta - pseudozeta - mu - pseudoalpha-1 - alpha-2 - alpha-1 - theta - 3'. The alpha-2 (HBA2) and alpha-1 (HBA1) coding sequences are identical. These genes differ slightly over the 5' untranslated regions and the introns, but they differ significantly over the 3' untranslated regions. Two alpha chains plus two beta chains constitute HbA, which in normal adult life comprises about 97% of the total hemoglobin; alpha chains combine with delta chains to constitute HbA-2, which with HbF (fetal hemoglobin) makes up the remaining 3% of adult hemoglobin. Alpha thalassemias result from deletions of each of the alpha genes as well as deletions of both HBA2 and HBA1; some nondeletion alpha thalassemias have also been reported.

圖像	Overlay histogram showing K562 cells stained with APR06167G (green line). The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were then icubated in 2% bovine serum albumin to block non-specific protein-protein interactions followed by the antibody (APR06167G, 1:25 dilution) for 60 min at 37?C. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight? 488 Conjugated Highly Cross-Adsorbed(OH191631) at 1/200 dilution for 40 min at 37?C. Isotype control antibody (blue line) was rabbit IgG (1μg/1x10^6 cells) used under the same conditions. Acquisition of >10, 000 events was performed. Anti-HBA2 Antibody (Center) at 1:2000 dilution + human fetal heart lysate Lysates/proteins at 20 μg per lane. Secondary Goat Anti-Rabbit IgG, (H+L), Peroxidase conjugated at 1/10000 dilution. Predicted band size : 15 kDa Blocking/Dilution buffer: 5% NFDM/TBST. Western blot analysis of HBA2 Antibody (Center) (Cat. APR06167G) in mouse liver tissue lysates (35ug/lane). HBA2 (arrow) was detected using the purified Pab.
